Question about cloning

    Default Question about cloning

    Hi everyone, when you do your cloning, do you use vented lids for that? And do you keep the flasks in a sealed space, like when you're doing your flasking? I need your ideas because that's gonna be my next project this weekend when my two special orchids arrived. Thank you.

    It's not necessary to use vented lids. It was a preference but most commercial growers use it as it has other advantages. were the filtration discs easily obtained in your area i would encourage you to use.

    while carrying out flasking it has to be done in sterile chamber ...using an inexpensive plexiglass box or glass aquarium turn over with the opening towards you OR a more costly setup using Laminar Flow Hood.

    I am a starter too but i hope i'm answering your question.

    Rob, when u sterillize ur specimen for tissue culture what do u use, bleach solution or H2O2?

    I use bleach ( sodium hypochlorite or commonly known as chlorox used at home) as H202 ( hydrogen peroxide) are not sold here, or was it ban i'm not sure cos some years ago it was easily bought. H202 will work too.

    For bleach i prepared 5% in distill water and sterilise 5 minutes.

    Using H202, bleach or ethanol has margin of error and you may vary the time 4 minutes and 3 minutes for different flasks.
