In-vitro culture of flower stalk internodes of phalanopsis

Hallo,

I have some orchids from wich I want to try to culture the flower stalk internodes. I have tried this before without any succes. The problem was not contamination, but just nothing happening at all. Is there anybody who did this already with succes, who want to share the secrect of getting PLB's?

I have been trying with P668 with:
BA 1mg/l ;
BA 5mg/l ;
BA 1mg/l + NAA 0.5mg/l ;
BA 5mg/l + NAA 0.5mg/l ;

No succes!

Are the lateral meristems viable? You might want to try before the flower stalk matures (maybe like when 6-12 inches in length). You might also wait for a couple months after flowering to harvest the meristems. This way the regulatory pathway for flowering might have weakened and the meristems might be more responsive to hormones.

A couple other quick things:
Do you think that you might have over sterilized the tissue?
Are you doing liquid culture and if so, are they getting enough aeration?
How are you dissecting out the meristems?
You might just have to try multiple times, unfortunately.
